Recombinant Human LR3-IGF-1 (Long Arg3 Insulin-like Growth Factor-1) is an engineered long-acting analog of human insulin-like growth factor 1, produced in an E. coli expression system in a tag-free form. Based on the native IGF-1 sequence, this protein carries a site-directed mutation (glutamic acid at position 3 replaced by arginine) and a 13-amino acid extension at the N-terminus, which significantly reduces its binding affinity to IGF binding proteins (IGFBPs), thereby greatly improving in vitro and in vivo bioavailability and functional half-life, with markedly stronger proliferation-promoting activity than native IGF-1. LR3-IGF-1 binds to and activates the IGF-1R receptor, triggering downstream signaling pathways such as PI3K/Akt and MAPK, and potently promotes cell proliferation, protein synthesis and cell survival. It serves as a core growth factor in serum-free cell culture, organoid expansion, stem cell maintenance and muscle development research.
